I think you'd have to let it dry before rolling it back up.

The ink would immediately run everywhere when you're drawing it. It's not about letting it dry before you roll it back up. The ink bleed would happen instantly once the marker touches the toilet paper. Toilet paper and a marker would be a bad combination for such a drawing.
